ANRNew funding of ANR JCJC 2019

The REGOS team of the INSERM U1229-RMeS laboratory is pleased to announce the success of the BILIPOX project in ANR JCJC 2019. The laboratory will receive a grant of 305k€ for 3 years to hire dedicated staff (Engineer and technician). The BILIPOX project will be coordinated by Alexis Gaudin and will be carried out in collaboration with Christophe Blanquart (CRCINA, INSERM UMR 1232), Philippe Legrand (UMR 5253 CNRS- UM-ENSCM), Jeff Johnson (Cayman Chemical Company) and Stéphane Simon (University Paris Diderot (Paris VII). The specific goal of this project is to evaluate the potential benefits of a new biphasic microgel as lipoxin A4 (LXA4) delivery system for tissue engineering (pulp and periodontal tissue). The results of BILIPOX may provide a better understanding of microgels/macrophage interaction. This would be an important milestone for hydrogels modular strategy that open a new way to design and to multi-functionalize drug delivery systems.

H2020An additional Horizon 2020 European research grant for INSERM UMR 1229-RMeS towards treating chronic back pain

In 2016, the European commission invested 5.5 millions Euros in the RESPINE project to clinically address whether bone marrow-derived mesenchymal stromal cells may help solve low back pain that affects hundreds of millions of patients worldwide. The double blind phase 2b RESPINE clinical trial (P.I. Christian Jorgensen, Montpellier University Hospital; EudraCT 2017-002092-25) is now recruiting more than 100 patients in France and European countries. Jerome Guicheux (Nantes scientific coordinator, STEP Team, INSERM UMR 1229-RMeS) is a task leader in this project in charge of developing cutting-edge potency assays for the preclinical evaluation of the regenerative potential of stem cells (Cécile Boyer). Fifteen patients will be recruited at the Nantes University Hospital in the department of neurotraumatology and neurosurgery (Eric Bord). After the successful funding of RESPINE, the STEP team of the INSERM UMRS 1229-RMeS is now pleased to announce the recent selection of the H2020 iPSpine project by the European commission. iPSpine will receive an overall grant of 15 M€. This large European consortium will gather 20 international partners and will be coordinated by Marianna Tryfonidou (Utrecht University, The Netherlands). This project will officially be launched in February 2019 and it will deal with the development of advanced therapy medicinal products based on the use of biomaterials and induced-pluripotent stem cells (iPSC) towards solving intervertebral disc (IVD) degeneration. Jerome Guicheux is the scientific coordinator of iPSpine workpackage (WP) 1 “iPSC development, differentiation and technologies” that gathers 7 international partners and focus on discovering innovative culture protocols for the generation of IVD cells derived from human iPSC (Anne Camus). In collaboration with the ONIRIS-national veterinary school of Nantes, the STEP team will also participate in WPs 3 and 6 respectively dedicated to (i) in vitro 3D cell culture (Catherine Le Visage, RMeS) and (ii) the first canine clinical trial related to the efficacy of the intradiscal injection of iPSC-derived regenerative cells in the treatment of low back pain (Marion Fusellier & Johann Clouet). RMeS will receive a global support of about 1.3 million Euros and will recruit 1 post-doc, 2 research assistants and 1 clinical assistant.

The STEP team is pleased to announce the funding of Vianney Delplace’s position by the Fondation pour la Recherche Médicale (FRM) for 3 years. Using his expertise in hydrogel design, Vianney Delplace will develop innovative intra-articular cell therapies for the treatment of osteoarthritis, under the supervision of Catherine Le Visage and Jérôme Guicheux. In collaboration with Arnaud Tessier from the CEISAM laboratory (UMR CNRS 6230, Nantes), a new class of self-healing hydrogels will be designed to best mimic the biomechanical properties of native cartilages and deliver anti-inflammatory factor secreting stem cells.

A triple success for the INSERM-Nantes University-ONIRIS UMRS 1229-RMeS laboratory at the 2018 call for proposal of the French national research Agency (ANR) as coordinator. The STEP team is pleased to announce the funding of two novel projects, PPAROA and KLOTHOA. PPAROA is an ANR collaborative research project (PRC) that will be coordinated by Jerome Guicheux (INSERM UMRS 1229-RMeS). PPAROA will aim to define the role of the nuclear Peroxisome Proliferator-Activated Receptor (PPAR) beta/delta in the anti-osteoarthritic and immunomodulatory properties of adult mesenchymal stem cells. In partnership with the Christian Jorgensen’s Laboratory (INSERM UMRS 1183-IRMB, University of Montpellier), the PPAROA project will be supported by the ANR for 4 years. KLOTHOA is an ANR young researchers (JC/JC) project that will be coordinated by Claire Vinatier (INSERM UMRS 1229-RMeS). KLOTHOA will aim to define the role of the anti-aging protein Klotho in the onset of osteoarthritis and whether modulating the expression of Klotho by adeno-associated virus-mediated gene therapy may help identify novel anti-osteoarthritic treatments. The KLOTHOA project will be supported by the ANR for 3 years. The REGOS team is also pleased to announce the funding of an industrial collaborative research project. The IXBONE project, coordinated by Pierre Weiss (INSERM UMRS 1229-RMeS) will aim to define new bone regeneration strategies for the treatment of mandibular osteoradionecrosis using mesenchymal stem cells and innovative biomaterials. In partnership with the French Institute for Radiological Protection and Nuclear Safety (IRSN, Fontenay aux Roses, Noëlle Mathieu) and the French Organ, Tissue, Regeneration, Repair and Replacement company (OTR3, Paris, Denis Barritault), the IXBONE project will be supported by the ANR for 4 years.

The STEP team is pleased to announce the funding of the CAPSTEM project by the Fondation de l’Avenir pour la Recherche Médicale Appliquée. CAPSTEM will be coordinated by Catherine Le Visage in collaboration with Claire Vinatier and Jerome Guicheux from RMeS laboratory, Denis Renard from the BIA laboratory (INRA UR 1268, Nantes) and Anne Des Rieux from the Louvain Drug Research Institute (LDRI, Louvain, Belgium). CAPSTEM will aim to determine whether encapsulating mesenchymal stem cells in hydrogel-based microcapsules may help exploit their anti-osteoarthritic properties. The CAPSTEM project is funded for 2 years and will be conducted in the framework of Audrey Smith’s PhD Thesis.

NexTNexT Initiative Research Grant

The STEP team is pleased to announce the funding of the SHELBY project by the NEXT initiative of the Nantes University (Internal Interdisciplinary Projects). SHELBY will be coordinated by Catherine Le Visage in collaboration with Claire Vinatier and Jerome Guicheux from RMeS laboratory, Denis Renard from the BIA laboratory (INRA UR 1268, Nantes) and Arnaud Tessier from the CEISAM laboratory (UMR CNRS 6230, Nantes). SHELBY will aim at developing a new generation of functionalized boronic acid hydrogels with self-healing properties and evaluating their ability to encapsulate mesenchymal stem cells and prevent osteoarthritis. The SHELBY project is funded for 1 year with possible extension up to 4 years and will be developed in the framework of Fabien Nativel’s PhD Thesis.

Marie-Michèle is post-doc. She will work on 3D biomimetic 3D printing cement for personalized repair of alveolar cleft. She will work on printable formulation of calcium phosphate cement using hydrogels in collaboration with GPEA, LS2N and INRA. She is implicated in the bioregate 3D refentine program coordinated by Pierre Corre. The objective is building adapted 3D bone scaffold for children with cleft palate.

BioregateAward for GENOA project

We are pleased to announce that the GENOA project "Gene therapy for osteoarthritis: controlled expression of anti-geronic factors" headed by Claire Vinatier in the STEP team is laureate of the call for proposal 2018 of the BIOREGATE regenerative medicine cluster funded by the region Pays de la Loire. GENOA will receive a 3-year fellowship for a PhD student and substantial funds for covering scientific experiments as well as valorization and dissemination activities. GENOA will be conducted in collaboration with Oumeya Adjali, head of the translational gene therapy for genetic diseases laboratory (INSERM UMR 1089, Nantes). GENOA will aim at (i) deciphering the role of the anti-geronic factor KLOTHO in the onset of osteoarthritis (ii) determining whether the adeno-associated virus-mediated intraarticular overexpression of this protein may help control disease progression in murine models

Pauline Chichiricco received an award for the best oral presentation at the BPG Conference (Belgian polymer group) held on 28-29 May 2018 in Blankenberge, Belgium. We are pleased to announce that the PPAROA project "Improvement of the therapeutic effect of CSM in osteoarthritis: Role of PPAR b/d" headed by C. Vinatier & J. Guicheux in the framework of B. Bodic's PhD thesis is laureate of the call for proposal 2018 "research & transplantation " from the French biomedicine agency. This project, selected from 90 proposals, will receive 30,000 € and will be conducted in collaboration with the INSERM UMR 1183-IRMB (F. Djouad & C. Jorgensen, Montpellier).

Foreum The STEP team welcomes the selection of the SENOA project to identify the role of senescence in osteoarthritis and its potential targeting for the treatment of this disease. This project is supported for 600 K€ by the Foundation for Research in Rheumatology (FOREUM, Switzerland) of the European League Against Rheumatism (EULAR).
Claire Vinatier will be in charge of the coordination of the working group n ° 2 with the objective of characterizing senescence and the associated molecular mechanisms in osteoarthritic joints.
